Abstract

|
Obtaining chitosan oligosaccharide having higher molecular weight than the usually obtained oligosaccharides by enzymatic digestion of high molecular weight chitosan was performed in vitro using beta-cyclodextrin (beta-CD) as inhibitor for chitosanase hydrolysis activity. The concentration of beta-CD:chitosan was 5:1 respectively. Beta-CD is cyclic oligosaccharide consists of eight glucopyranose units linked by ¥á-1,4 glycosidic bonds. It was capable of inhibiting chitosanase activity to 59.2% less activity at 37¡É after 2 hr incubation and we got chitosan oligosaccharide with higher molecular weight that can be separated by HPLC. The antibacterial activity of this chitosan oligosaccharide was also investigated. By condensing the products of reaction mixture having 2.5% beta-CD+0.5% chitosan+chitosanase 5 times less volume to get final concentration of chitosan 2.5%, this sample demonstrated a remarkable antibacterial activity.
